| Citation | Gutfreund's early work on the molecular weights of proteins and the reversible association of insulin and of haemoglobin made a significant contribution to a difficult subject. His principal achievements have come from applications of rapid reaction techniques to the identification and characterisation of elementary steps in reactions catalysed by proteolytic enzymes, phosphate and dehydrogenases. He was the first to obtain direct estimates of the rate of substrate combination with proteases, and of the rate of hydrogen transfer from substrate to coenzyme in the catalytic step of a dehydrogenase reaction. He obtained kinetic evidence that conformational changes of the enzymes occur as distinct steps in the mechanisms of alkaline phosphatase and lactate dehydrogenase. Recent studies by Gutfreund and his collaborators of the kinetics of proton release in the alcohol and lactate dhydrogenase reactions have contributed substantially to our understanding of these aspects of the mechanisms. The research group set up by Gutfreund in Bristol is widely recognised as a productive centre for the application of rapid reaction techniques to the problems of enzyme mechanisms, and his publications, including two text books, have helped considerably to further work in this important field. |
