Citation | Has illuminated important aspects of interaction between DNA and proteins. He removed distinct fractions of histones from chromatin and showed that heterogeneity within one fraction arose from genetic polymorphism. He established methods for the fractionation and analysis of small fragments of DNA, thus laying the foundations for subsequent work on DNA sequences. He also developed methods based on terminally labelling DNA for determination of nucleotide sequences at sites of action of restriction and modification enzymes and ter systems of phages, particularly lambda. These sites involve rotationally symmetrical sequences specific for each enzyme, some restriction enzymes leaving cohesive ends. This led to the development of phage lambda as a receptor chromosome for DNA fragments generated by appropriate restriction enzymes. The technique permits heterologous recombination between DNAs from widely ranging sources and promises to be of great fundamental and practical significance. |
