| Citation | Howard Temin is one of the most eminent virologists. His career has been dedicated to unravelling the mechanism of replication of retroviruses and he is distinguished as the proposer of a DNA intermediate in this process and as the discoverer of reverse transcriptase. His early studies of cell transformation by Rous sarcoma virus and of the cell growth controlling factors in serum led to considerations of the mechanism of viral carcinogenesis in which he developed the concepts of the synthesis of viral specific DNA sequences during infection and their integration into the cellular chromosome. His discovery of the virion reverse transcriptase of Rous sarcoma virus with its subsequent impact on molecular biology followed directly from this work and he awarded the Nobel Prize for Medicine or Physiology in 1975. More recently the development of retroviruses as vectors, the discovery of the similarities between retroviral and long terminal repeat sequences and those of mobile genetic elements and the discovery of encapsidation and integration sequences in retrovirus genomes are fundamentally important contributions to virus biochemistry and molecular genetics. Moreover, all this information is likely to be of considerable practical significance in designing drugs for the chemotherapy of retrovirus infections. |
