Citation | Randle has defined the enzymic steps mediating the Pasteur effect in muscles and discovered the mechanism whereby lipid fuels inhibit glucose utilization and contribute to secondary manifestations of insulin deficiency. He has discovered mechanisms whereby insulin deficiency leads to sustained phosphorylation and inactivation of pyruvate dehydrogenase, pioneered the use of halogenated carboxylic acid inhibitors of pyruvate dehydrogenase phosphorylation and has thereby shown the importance of this phosphorylation of the enzyme complex in glucose conservation; this has provided a therapeutic method of removing blood and tissue lactic acid. That insulin secretion was specifically stimulated by a variety of sugars and was inhibited by adrenaline was discovered by Randle. He also discovered, and elucidated the mechanism of, mannoheptulose diabetes and originated a beta-cell glucoreceptor mechanismbased on glucose phosphorylation. |
