Citation | Patrick Chinnery has advanced our understanding of how mitochondrial diseases originate, is inherited and expressed. He showed that a reduction in mitochondrial DNA (mtDNA) content within primordial germ cells leads to major changes in mutation load over one generation. This explains how mutations spread rapidly throughout the population and cause diseases of differing severity. He showed that most humans harbour different mixtures of mtDNA molecules which provides a reservoir for population genetic diversity which is modulated by the cell nucleus during maternal transmission, and that there is ongoing transfer of mtDNA into the cell nucleus shaping genome evolution. |
