Citation | Shi’s landmark discovery of the first histone demethylase, LSD1, overturned the long-held dogma that histone methylation was irreversible and set the standard for an avalanche of additional demethylase discoveries for which Shi was a major contributor. Shi and collaborators provided the first atomic level insight into mechanism of demethylation and transcriptional regulation. Shi discovered the link between histone methylation dynamics and intellectual disability and developed strategies, now in clinical trials, to enable immune checkpoint blockade by modulating LSD1. Shi’s extraordinary record of pioneering/paradigm-shifting discoveries have revolutionized the fields of chromatin and epigenetics and have far-reaching impacts in biology and medicine. |
