Citation | BV has made fundamental discoveries about how lipid second messengers are regulated to control cellular and organismal functions. His contributions include showing non-redundant functions of different phosphoinositide-3- kinase (PI3K) isoforms. Notably, he identified and characterised PI3K-p110 which is expressed selectively in leukocytes and some cancer cells. It is now known that mutants of this enzyme cause human immunodeficiency and autoimmunity. He has driven the generation of isoform-selective PI3K-inhibitors that are used clinically as immune therapeutics and for cancer treatment. His recent development of small-molecule PI3K activators has turned decades of PI3K research on its head, opening new avenues for kinase drug development. |
