Citation | Distinguished for his work on the proteins of sarcoplasmic reticulum involved in release, storage and uptake of calcium ions. This has involved purification, biochemical characterisation, cloning and sequencing of six different proteins several of which he identified for the first time. His major achievement has been a structural model for the calcium pump (with N. M. Green) based upon its amino acid sequence and the properties of over 250 site-directed mutants. Study of these mutants led to new insights into the mechanism of the pump, especially from his identification of the cation transport sites within the membrane. He has shown that the genetic defect in malignant hyperthermia is the consiquence of a mutation at a single site in the 5,000 residue sequence of the calcium release channel, which he had previously cloned and sequenced. |
