Citation | Pawson is distinguished for his outstanding contributions to our understanding of the mechanisms by which cells respond to external signals generated by the interaction of growth factors with receptors which trigger cellular growth and differentiation. Through biochemical and genetic studies of the transforming proteins of certain oncogenic retroviruses, he discovered the src homnolgy 2 domain (SH2), proposed that this protein module was involved in the interaction of tyrosine kinases with their substrates, and demonstrated that SH2 domains interact with the phosphotyrosine residues of ligand-activated receptor tyrosine kinases. He also recently defined the mechanism by which tyrosine kinase receptors activate the proto oncogene ras, the first description of a physiological interaction involving src homology region 3 (SH3) domains. Pawson's studies of proteins with SH2 and SH3 domains, which have been made in a highly competitive area, have dramatically advanced our knowledge of the complex networks involved in cellular regulation. His discoveries are of considerable general significance for the actions of growth factors and oncogenes in view of the large number of proteins with SH2 and SH3 domain that are now being identified and the innumerable biological responses which they regulate |
