Citation | Harren Jhoti is recognised internationally for his innovative use of structural biology and fragment-based approaches for drug discovery, first in GlaxoWellcome and then Astex, which he co-founded in 1999. He pioneered concepts widely adopted by Pharma, such as the 'Rule-of-Three', which now underpins most work in the field. Jhoti's group solved the first structures of drug complexes with human cytochrome P450 isoforms, a major advance for design of compounds more stable towards these drug-metabolising enzymes. His fragment approach has been particularly successful for oncology targets leading to eight drugs entering clinical trials, with four licensed by major pharmaceutical companies and one on the market for breast cancer. |
