Citation | Ian Tomlinson is a clinician scientist who has combined molecular genetic analysis of familial and sporadic cancer with population studies to identify new mechanisms of cancer growth and new theories of cancer evolution. He has discovered novel and unexpected oncogenic pathways including dysregulated bone morphogenetic protein signalling in colorectal cancer and defects in the Krebs cycle enzyme, fumarate hydratase in kidney cancer. Through precise analyses of the mutational spectrum in cancer and the functional consequences, his work has also changed general thinking towards a "just right" model in which tumourigenesis is driven by quantitative functional changes that are neither too weak nor too strong. |
