Citation | Scheres is best known for two principle innovations in macromolecular structure determination using electron cryomicroscopy. First, he developed 3D classification as a novel approach to structure analysis, revealing hidden multiple states within single cryoEM data sets. Second, he created a key tool that has been critical to the recent cryoEM revolution in structural biology, and which has resulted in the determination of 3D structures of hundreds of previously intractable biological macromolecules. He implemented Bayesian statistical methods in his RELION image-processing package, and this has produced such a powerful improvement that during the last few years the vast majority of recent structure determinations have used his methods. Scheres has applied his new methods to the determination of many key biological structures, including the ribosome, spliceosome, gamma-secretase and tau amyloid filaments. |
