Citation | Distinguished for research on the chemistry and biochemistry of nucleic acids. In 1952, by showing that isomeric ribonucleotides have a behaviour parallel to the glycerophosphates, Brown was able to reformulate a 3'-5' polynucleotide structure for RNA which accounted for its hydrolytic behaviour. This seminal paper also had recognised implications for the structure of DNA and it led directly to procedures for determining the structures of oligonucleotides, and to the understanding of the mechanisms of ribonuclease action. A considerable amount of synthetic work was done to prove the consequences of the models proposed. Brown has also made contributions to other aspects of nucleotide and also phospholipid chemistry. In more recent years he turned his attention to the chemical basis of mutagenesis by reagents such as hydroxylamine, using in vitro replication of polynucleotides. His work on the chemical modification of transfer RNA was important in the understanding of the 3-dimensional structure of this molecule. |
