Citation | Distinguished for his contributions to our understanding of cellular immunology, with particular reference to non-specific immunity, graft-versus-host (GVH) reactions and the mechanism of tolerance induction. Howard's early work on bacterial immunity foreshadowed much recent work on the enhancement of non-specific immunity by bacterial lipopolysaccharides and lipid A. He has maintained a long interest in adjuvants, and particularly C.Parvum as a potentially useful agent for the enhancement of immune responses. In his study of GVH reactions, he defined the accompanying immunosuppression and macrophage activation. Howard's main body of work has been directed at tolerance. Early studies revealed that induction of transplantation tolerance in the new-born reflects antigen dose rather than a qualitative difference from the adult. In recent years, an extensive survey of polysaccharides and their structural relationship to tolerogenicity supports the concept that B-cell tolerance to thymus-independent antigens reflects receptor modulation. The findings have possible practical applications for induction of tolerance in drug allergies. |
