CitationPaul Lehner is a pioneer in studying virus-host antagonism. Imaginatively applying unbiased quantitative proteomics to viral evasion he transformed our understanding of how human viruses remodel the infected cell landscape. Using genetic screens, he discovered the ‘HUSH’ epigenetic silencing complex that discriminates newly-introduced retroelements from endogenous genes, uncovering a novel transcription-dependent, vertebrate genome surveillance system that protects the genome from integrating retroviruses (e.g. HIV) and retrotransposons. HUSH’s unique ability to differentiate self from non-self genomic DNA, through recognition of intronless DNA, provides unanticipated mechanistic insight into self-recognition at the genomic level, paving the way to applications in medicine and biotechnology.
